If you have 6 cards in your hand and you have tsukuyomi out in the field. Do you have to discard or not when you end your turn?

Your turn cannot end if you have more than 6 cards in your hand, if you end your turn and a spirt monster is face-up, it will return to your hand, and if you now hold more 6 cards in your hand, you will still have to discard.

To be a bit more technical a spirit monster returning to the hand is something that must be resolved before you may leave the End Step. The discarding to get down to 6 cards happens at the very end of the End Step. Regardless any effect which would add an additional card to your hand while you are still completing the "discard to get down to 6" will cause you to discard again.
f you have "Forced Requisition" active (face-up) and 7 cards in your hand during the End Phase (and one of them is a "Night Assailant"), and there is another "Night Assailant" in your Graveyard, and you discard your "Night Assailant" because you have 7 cards, your opponent must discard because of "Forced Requisition". Then you can activate the effect of "Night Assailant" to retrieve the other "Night Assailant". Now you have 7 cards in your hand again so you can discard "Night Assailant", and your opponent must discard again for "Forced Requisition". You can continue doing this until your opponent loses all cards in their hand.
